Best APC2 Antibody: How to Identify High-Quality Products
The cellular mechanisms of APC2 action are not fully understood. We recently reported that the cytoplasmic domain of APC2 is acetylated, and this acetylation is required for APC2 interaction with microtubules. We therefore examined whether APC2 overexpression alters microtubule dynamics by transfecting L cells and B103 cells with a control or APC2 expression plasmid, and then analyzing cell structures using anti-acetylated or anti-detyrosinated tubulin antibodies. Overexpression of APC2 significantly enhanced the colocalization of acetylated tubulin, and a 2.5-fold or 2-fold increase in the concentration of acetylated tubulin was observed when APC2 expression was increased.
We also performed ephrin-A2 stripe assays with control and APC2 knockdown nasal and temporal axons in cultured hippocampal neurons and found that both the control and APC2 knockdown axons avoided ephrin-A2-Fc stripes, but the APC2 shRNA-treated nasal axons did not (Fig. 7A). Nocodazole treatment, which disruptes microtubule stability, reduced the avoidance of ephrin-A2-Fc by both the control and APC2 knockdown nasal axons. The results suggest that APC2 functions as an MAP-like protein in neurons by stabilizing microtubules, and this function is essential for the normal response of retinal axons to a repulsive guidance molecule in the tectum.
